RESUMO

ObjectiveTo observe the effects of Hedysari Radix polysaccharide on the apoptosis of gastric sinus smooth muscle cells and explore the underlying mechanism via the insulin-like growth factor-1 (IGF-1)/phosphatidylinositol 3-kinase (PI3K)/serine-threonine kinase (Akt) pathway in the rat model of diabetic gastroparesis (DGP). MethodSixty-two Wistar male rats were randomized into a blank group (n=12) and a modelling group (n=50). The rat model of DGP was established by small-dose multiple intraperitoneal injections of streptozotocin combined with an irregular high-fat and high-sugar diet for 4 weeks. The modeled rats were randomized into model group, mosapride citrate (1.35 mg·kg-1), and high-, medium-, and low-dose (200, 100, and 50 mg·kg-1, respectively) Hedysari Radix polysaccharide groups. The rats were administrated with corresponding drugs by gavage, and those in the blank and model groups with equal volumes of pure water by gavage once a day for 8 consecutive weeks. The random blood glucose and body mass were measured every 2 weeks, and gastric emptying rate was calculated. Hematoxylin-eosin (HE) staining was used to observe the pathological changes of smooth muscle in gastric antrum, and terminal deoxynucleoitidyl transferase-mediated nick-end labeling (TUNEL) was employed to detect the apoptosis of smooth muscle cells in the gastric antrum. The expression of IGF-1, phosphorylated (p)-PI3K, and p-Akt in the smooth muscle of gastric sinus tissue was detected by immunohistochemistry. Western blot was employed to determine the protein levels of IGF-1, p-PI3K/PI3K, p-Akt/Akt, B-cell lymphoma-2 (Bcl-2), and Bcl-2-associated X protein (Bax) in the smooth muscle of the gastric antrum. ResultCompared with the blank group, the model group showed elevated random blood glucose at all time points (P<0.01), decreased body mass and gastric emptying rate (P<0.01), increased apoptotic index of smooth muscle cells in the gastric antrum (P<0.01), down-regulated protein levels of IGF-1, p-PI3K/PI3K, p-Akt/Akt, and Bcl-2, and up-regulated protein level of Bax (P<0.01). Compared with the model group, the 8 weeks of drug administration lowered the random blood glucose, increased the body mass and gastric emptying rate (P<0.05, P<0.01), decreased the apoptotic index of smooth muscle cells in the gastric antrum (P<0.05, P<0.01), up-regulated the protein levels of IGF-1, p-PI3K/PI3K, p-Akt/Akt, and Bcl-2, and down-regulated the protein level of Bax (P<0.05, P<0.01). Compared with the mosapride citrate group,the administration of low-dose Hedysari Radix polysaccharide for 6 and 8 weeks lowered the random blood glucose and decreased the body mass (P<0.05, P<0.01),low and medium-dose Hedysari Radix polysaccharide decreased the gastric emptying rate and the apoptotic index of smooth muscle cells in the astragaloside low-dose group decreased (P<0.05). The protein levels of IGF-1,p-PI3K/PI3K,p-Akt/Akt and Bcl-2(low dose)were down-regulated and the protein level of Bax was up-regulated by low doses of Hedysari Radix polysaccharide (P<0.05, P<0.01). Compared with high-dose Hedysari Radix polysaccharide, low-dose Hedysari Radix polysaccharide elevated random blood glucose and reduced body mass after 6 and 8 weeks of administration (P<0.05, P<0.01), and the low and medium doses decreased the gastric emptying rate, increased the apoptotic index of smooth muscle cells in the gastric antrum (P<0.05, P<0.01), down-regulated the protein levels of IGF-1, p-PI3K/PI3K, p-Akt/Akt, and Bcl-2, and up-regulated the protein level of Bax (P<0.05, P<0.01). Compared with the medium-dose group,the low-dose group of Hedysari Radix polysaccharide had lower body mass,lower gastric emptying rate in rats,higher apoptotic index of smooth muscle cells in gastric sinus tissue after 6 and 8 weeks of administration (P<0.05, P<0.01), and lower protein expression of IGF-1,p-PI3K/PI3K,p-Akt/Akt. ConclusionHedysari Radix polysaccharide protects the smooth muscle cells in gastric antrum against apoptotic injury and promotes gastric motility by activating the IGF-1/PI3K/Akt signaling pathway, as manifested by the up-regulated expression of IGF-1, p-PI3K, p-Akt, and Bcl-2 and down-regulated expression of Bax.

Objective:To explore the method and experience of augmentation mammoplasty with autologous fat transplantation assisted by external expander.Methods:From January 2013 to December 2020, 352 female patients (698 breasts, 346 bilateral and 6 unilateral), aged from 18 to 55 years, with an average age of 41.5 years, were admitted to the Department of Cosmetic Surgery, Beijing Jingmei Medical Cosmetology Clinic. From 1-28 days before surgery, autologous fat transplantation was performed after wearing the dilator, and postoperative wearing of the dilator lasted from 1 to 24 months, with an average of 3.5 months.Results:The volume of fat transplantation per breast ranged from 150 ml to 460 ml, with an average of 320 ml. No obvious postoperative complications such as fat liquefaction, infection, intramammary induration and fat embolism were observed, and the postoperative appearance was lasting. The postoperative follow-up was conducted from 6 months to 7 years, and patient postoperative satisfaction rate reached 84.9% and doctors' satisfaction after surgery reached 82.1%.Conclusions:Augmentation mammoplasty with autologous fat transplantation assisted by external expander can effectively improve the appearance of breast morphology and contour defects and increase the survival rate of fat transplantation, which is worthy of clinical application.

In the past 37 years, human immunodeficiency virus/acquired immunodeficiency syndrome (HIV/AIDS) has undergone various major transmission routes in China, with the world most complex co-circulating HIV-1 subtypes, even the prevalence is still low. In response to the first epidemic outbreak of HIV in injecting drug users and the second one by illegal commercial blood collection, China issued the Anti-Drug Law and launched the Blood Donation Act and nationwide nucleic acid testing, which has avoided 98,232 to 211,200 estimated infections and almost ended the blood product-related infection. China has been providing free antiretroviral therapy (ART) since 2003, which covered >80% of the identified patients and achieved a viral suppression rate of 91%. To bend the curve of increasing the disease burden of HIV and finally end the epidemic, China should consider constraining HIV spread through sexual transmission, narrowing the gaps in identifying HIV cases, and the long-term effectiveness and safety of ART in the future.

Objective: To improve the time window of early diagnosis of chronic obstructive pulmonary disease (COPD) and make quantitative evaluation of lung function using the voxel-based quantitative CT. Methods: Early COPD and COPD patients who had received inspiratory and expiratory CT scanning were consecutively recruited from the multi-center study named Digital Lung. Quantitative parameters of functional small-airway disease (fSAD) and emphysema (Emph) were measured. For the all COPD patients, Pearson correlation analysis or Spearman rank correlation analysis was used to test the correlation between the quantitative measurements of CT and pulmonary function. Results: Finally, 37 patients with early COPD and 50 patients with COPD were included. The mean value of Emph in early COPD was (3.50±3.36)% and the average value of fSAD was (18.12±12.73)%. Meanwhile, the mean value of Emph in COPD patients was (13.46±9.03)% and the mean value of fSAD was (36.57±9.88)%. Emph was negatively correlated with FEV1% and FEV1/FVC (r=-0.470, P=0.001; r=-0.334, P=0.018), respectively. fSAD was negatively correlated with FEV1% and FEV1/FVC (r=-0.453, P=0.001; r=-0.320, P=0.001). Conclusion: The voxel-based quantitative CT is helpful in early detection of small airway injury in early COPD with respiratory symptoms but no pulmonary function can be achieved.

<p><b>OBJECTIVE</b>To investigate the reporting characteristics of case reports of acupuncture therapy with CAse Report (CARE) guidelines, and to explore the applicability of the guidelines for these case reports.</p><p><b>METHODS</b>Case reports published from January 1, 2011 to December 31, 2013, were identified by searching PubMed and the China Biomedicine Database by using MeSH terms. Screening and data extraction of case reports were conducted by two independent researchers. Assessments based on CARE guidelines and data analysis were conducted by using Excel 2003 and RevMan 5.0.</p><p><b>RESULTS</b>A total of 61 case reports published in 19 journals were identified, which involved 16 major types of diseases. Fifteen (24.6%) case reports declared no conflict of interests, and 5 (8.2%) reported funding sources. None of these case reports met all items of CARE guidelines, and only 6 subordinate items were fully reported among 50% of the case reports. Subgroup analysis indicated that case reports published in English or those that did not have competing interests had higher reporting quality.</p><p><b>CONCLUSIONS</b>The reporting quality of case reports of acupuncture therapy is rather low. The reason might be a lack of awareness among authors and the poor dissemination of information regarding CARE guidelines in China. More evaluation studies are needed to promote and improve adherence to CARE guidelines. Moreover, an extended version of CARE guidelines should be developed for cases reports of special interventions such as acupuncture or adverse events caused by interventions.</p>

The skeletal muscle atrophy could be induced by the injury of nerve. According to the source of denervated skeletal mus-cle atrophy, it could be divided into exogenous muscle atrophy and endogenous muscle atrophy. In recent years, the ex-ogenous muscle atrophy models are mainly established by operating, physically injuring or chemically injuring, while the endogenous muscle atrophy models are mainly established by the transgenic animals of amyotrophic lateral sclero-sis. The selection and optimazation of animal models are crucial for the basic studies of denervated skeletal muscle atro-phy.

Learning objective clarity is very important for college students' classroom learning . Health assessment course as one of the compulsory course of nursing teaching, is the basis of nursing students' learn nursing profession, therefore to explore the related factors of health assessment course learning objec-tives has its practical significance. Under the guidance of the concept of the reform of the teaching system, the SIMPP analysis of the students learning objective in the college nursing students in health assessment course has been carried out. Results show that among college nursing students, three kinds of students' learning goals are very clear, four kinds of students' learning goals are clear, and two kinds of students' learning goals are unclear. The related factors influencing the nursing students' learning goal mainly are the student personal subjective factors. Teachers should pay attention to the observation of the students' study status in health assessment course, understand the student individual situation, control the clear degree of learning goals among students and do the ideological work of students at the beginning of the course to im-prove the students' clear degree of learning goals.

RESUMO

Objective To evaluate the application of MSCT 3D reconstruction technology without contrast on pulmonary arterio-venous malformation(PAVM).Methods The clinical and imaging data of 12 cases with PAVM confirmed by DSA were analyzed ret-rospectively.The plain scan with 16 slice spiral CT scanner performed pulmonary 3D reconstruction at post-processing work station by methods of MPR,MIP and VR.Results All cases could be diagnosed accurately,and images could clearly show the spatial struc-tures and its relations of the feeding arteries,draining vein and aneurysmal sac.Conclusion MSCT post-processing technique of PAVM without contrast is able to make a precise diagnosis,and to show the configuration of PAVM,which does not need contrast medium and acquirelow radiation dose.

<p><b>OBJECTIVE</b>To investigate the value of psychological intervention in treating slow transit constipation (STC), and to provide the reference to clinical treatment for STC patients with psychological disorder.</p><p><b>METHODS</b>A total of 94 STC patients with psychological disorder admitted to the Anorectal Hospital of Chengdu from June 2010 to August 2012 were prospectively enrolled and divided into psychological intervention group(subtotal colectomy plus postoperative psychological intervention) and control group (subtotal colectomy without postoperative psychological intervention). Scores of Hamilton depression scale (HAMD), Hamilton anxiety scale(HAMA), Wexner constipation scale (WCS) and gastrointestinal quality-of-life index(GIQLI) were recorded 1, 3, 6, 12 and 24 months after operation. SPSS 17.0 statistical software was used to analyze the data.</p><p><b>RESULTS</b>There were no differences in baseline data, operative time, blood loss, time to the first flatus and time to the first defecation between two groups(all P>0.05). The scores of HAMD and HAMA were significantly reduced in psychological intervention group compared with control group 3, 6, 12 and 24 months after operation (all P<0.05). Ratios of cure, obvious progress, progress and invalidation of depression symptoms in psychological intervention group were 2.6%(1/39), 66.7%(26/39), 25.6%(10/39) and 5.1% (2/39) respectively at postoperative 24-month, which were better than those [0, 34.2%(13/38), 44.7% (17/38) and 21.1%(8/38) respectively] in control group with significant difference(P=0.013). Ratios of cure, obvious progress, progress and invalidation of anxiety symptoms in psychological intervention group were 10.3%(4/39), 53.8%(21/39), 28.2%(11/39) and 7.7%(3/39) respectively at postoperative 24-month, which were better than those [0, 28.9%(11/38), 55.3%(21/38) and 15.8%(6/38) respectively] in control group with significant difference (P=0.011). The WCS scores in psychological intervention group were lower than those in control group 6, 12, 24 months after operation (all P<0.05). The GIQLI scores in psychological intervention group were higher than those in control group 3, 6, 12 and 24 months after operation(all P<0.05).</p><p><b>CONCLUSION</b>Subtotal colectomy combined with psychological intervention not only can significantly improve psychological disorder, but also increase the efficacy of surgery in the treatment of slow transit constipation patients with psychological disorder.</p>

<p><b>OBJECTIVE</b>To evaluate and compare the value of dynamic multiple pelvic angiography and pelvic four-contrast defecography in the diagnosis of functional defecation disorder.</p><p><b>METHODS</b>From September 2014 to July 2015, a prospective controlled trial was carried out in Chengdu Anorectal Hospital. A total of 32 patients met the inclusion criteria of functional defecation disorder simultaneously underwent pelvic four-contrast defecography and dynamic multiple pelvic angiography. The diagnostic results of these two methods were compared.</p><p><b>RESULTS</b>The absolute values of anorectal angle and level of perineum, peritoneum and bladder from rest to defecation were (29.6±13.6)°, (26.2±14.2) mm, (55.5±25.6) mm and (28.9±16.5) mm in dynamic multiple pelvic angiography, and (24.6±5.8)° (18.7±10.6) mm, (34.5±18.4) mm and (19.2±11.8) mm in pelvic four-contrast defecography respectively, whose differences were statistically significant (P = 0.026, 0.022, 0.000, 0.011 respectively). The diagnostic rate of pelvic peritoneal hernia was 93.8%(30/32) and 68.8%(22/32) in dynamic multiple pelvic angiography and pelvic four-contrast defecography respectively with significant difference(P=0.011).</p><p><b>CONCLUSION</b>Dynamic multiple pelvic angiography has significant advantage in the diagnosis of pelvic peritoneal hernia, and can provide a more objective basis for the diagnosis of functional defecation disorder.</p>

Objective:To explore the quality on 64-slice CT coronary angiography with different injection rates of contrast agent.Methods: 120 patients who have been diagnosed or suspected coronary atherosclerosis were selected in our hospital and were randomly divided into group A(4.5ml/s), group B(5.0ml/s) and group C(5.5ml/s), with 40 cases in each group. We observed CT attenuation values, image quality, and adverse reactions of the three groups.Results: There were significant differences in the ascending aorta, descending aorta, left main, left anterior descending artery, circumflex artery and the right coronary artery and other parts of the CT attenuation value (F=6.838,F=4.002,F=7.261,F=5.992,F=8.281,F=5.790,F=6.126;P<0.05). In image quality, the group B with 34 cases was significantly better than group A with 12 cases and group C with 20 cases (x2=9.632,x2=5.475;P<0.05). The rate of adverse reactions was 30%, 7.5% and 22.5% with 12 cases in group A, 3 cases in group B and 9 cases in group C. The rate in group B was significantly lower than in group A and group C (x2=12.985,x2=10.236;P<0.05).Conclusion: Through the application of the same concentration of the contrast agent and scan conditions to coronary angiography, we can get a good vascular enhancement and high quality imaging when the injection rate of contrast medium is 5ml/s, while incidence of adverse reactions may be reduced.

<p><b>OBJECTIVE</b>To investigate the clinical application of laparoscopy-assisted subtotal colectomy with transanal specimen extraction for slow transit constipation(STC).</p><p><b>METHODS</b>Retrospective analysis was performed on the clinical data of 8 cases with STC undergoing the procedure mentioned above from February to November 2013. Pre-and post-operative constipation was assessed using Wexner Constipation and Incontinence Scales, and quality of life was assessed using Gastrointestinal Quality of Life Index.</p><p><b>RESULTS</b>All the operations were completely successful without postoperative complications, such as intestinal fistula, pelvic infection, anastomotic stricture, intestinal obstruction. The Operative time was (287.6 ± 21.5) min, blood loss was (109.7 ± 41.1) ml, time to first flatus was (2.5 ± 0.9) d. The proportion of postoperative constipation symptom index improvement was(77.6 ± 8.3)%. Postoperative quality of life score was 97.3 ± 15.7, significantly higher than that before operation(P<0.05). Postoperative Wexner constipation score was 8.8 ± 3.7, significantly lower than that before operation.</p><p><b>CONCLUSION</b>Laparoscopy-assisted subtotal colectomy with transanal specimen extraction in the treatment of STC has good short-term efficacy with obvious improvement in quality of life.</p>

<p><b>OBJECTIVE</b>The aim of this study was to understand the rates of infection and spontaneous clearance on hepatitis C virus (HCV) among HIV-infected men who have sex with men (MSM) in China.</p><p><b>METHODS</b>HIV-infected MSM from Shenyang, Beijing, Changsha and Kunming were selected to take the HCV antibody and HVC-RNA tests, and then classified into 'under recovery (RNA-/Ab+) ' or being chronic (RNA+/Ab+). Information on demography, CD4(+) T cell, HIV viral load and liver biochemical indicators were also collected.</p><p><b>RESULTS</b>In 513 HIV-infected MSM, the positive rate of HCV antibody was 1.94% (10/513) and the positive rate of HVC-RNA was 0.77% (4/513). Five patients showed clearance of HVC-RNA, spontaneously. These 5 patients had low levels of both alanine transaminase (ALT) and aspartate aminotransferase (AST), and with high level of CD4(+) T cells, when compared to those who were not able to clear the HVC-RNA.</p><p><b>CONCLUSION</b>Currently, the HCV infection rate was still at quite low level among the HIV-infected MSM in China. Dynamic of HIV/HCV co-infection should be monitored so as to develop suitable strategy on prevention and treatment among this population, in China.</p>

<p><b>OBJECTIVE</b>This study aimed to investigate the subtype distribution of gp41 gene of human immunodeficiency virus-1 (HIV-1) among men who have sex with men (MSM) in Zhengzhou.</p><p><b>METHODS</b>Thirty blood samples were collected from men who have sex with men infected by HIV. The complete gp41 gene was amplified by RT-PCR and nested-PCR and sequenced. All sequences were edited by Bioedit and subtyped with HIV sequence library US Los Alamos National Laboratory and online genotyping software provided by American National Center of Biotechnology Information. Phylogenetic analysis of gp41 gene was performed using the MEGA 3.1 software, and the genic dispersion rates among subtype of gp41 gene were analyzed.</p><p><b>RESULTS</b>A total of eighteen gene sequences of HIV-1 gp41 gene were obtained from thirty men who have sex with men infected by HIV, which belonged to subtype CRF15-01B (50% (9/18)), CRF01-AE (22% (4/18)), CRF07-B (22% (4/18)) and B (6% (1/18)), respectively. The intersubtype HIV-1 strains aggregate with according reference strains. The genetic distance inter-subtype of subtype CRF15-01B, CRF01-AE and CRF07-B were 0.050 ± 0.007, 0.052 ± 0.009 and 0.082 ± 0.012, respectively.</p><p><b>CONCLUSION</b>The prevalent subtypes of HIV-1 among among MSM in Zhengzhou was complicated and recombinant HIV-1 strains were the most prevalent strains.</p>

To investigate the subtype distribution of human immunodeficiency virus-1(HIV-1) infection among men who have sex with men (MSM) in Zhengzhou, Henan Province, forty blood samples were collected from HIV-1 carriers, who acknowledged to have sex with men. The complete gag gene was amplified by RT-PCR and nested-PCR and sequenced. All sequences were edited by BioEdit and subtyped by genotyping software. Phylogenetic analysis of gag gene were then performed using the MEGA 3.1 software, the gene distances were calculated by Distance program. There were three different HIV-1 subtypes including B, CRF01-AE and CRF07-BC present among twenty four MSMs in Zhengzhou. Genotyping results showed that 33.33% (8/24) were B, 41.67% (10/24) were CRF01-AE and 25% (6/24) were CRF07-BC, and subtype CRF01-AE had become the most prevalent HIV-1 subtype in Zhengzhou, Henan province. In conclusion, recombinant HIV-1 strains are circulating in Henan province and the epidemiology is complicated.

RESUMO

<p><b>OBJECTIVE</b>To observe the temperature change of traditional silver needle in the human body during the burning of moxa ball.</p><p><b>METHODS</b>Thirty-six healthy volunteers were randomly divided into a single-needle group and a multi-needle group, 18 cases in each group. For both groups, one silver needle (18 cm in length, 1.1 mm in diameter), which was adopted in this research to measure the temperature change, was punctured in the insertion point of the volunteer (inside the top of the left buttock, 7 cm under the edge of the highest point of the iliac crest, 7 cm lateral to the dorsomedian line), then another four silver needles were punctured 2 cm respectively anterior, posterior and lateral to the insertion point in the multi-needle group, and all the silver needles were inserted with 6 cm depth. Afterigniting the 1.3 g moxa ball on the needle tail, the temperature of the measuring points that were 3 mm, 33 mm, and 63 mm above the silver needle tip were recorded separately by digital temperature measuring instrument.</p><p><b>RESULTS</b>The peak temperature of the three measuring points in the single-needle group was all around 41 degrees C, while those in the multi-needle group were around 43 degrees C, which had significant differences (all P < 0.05), but no significant differences among the highest temperature of the measuring points in the same group could be found (all P > 0.05). The highest temperature of moxa ball in the single needle group was (611.16 +/- 6.91) degrees C, while that of the central moxa ball in the multi-needle group was (628.94 +/- 8.99) degrees C, the difference of which was significant difference (P < 0.01).</p><p><b>CONCLUSION</b>The temperature conductivity of the silver needle is very well, so the heat of the moxa ball could pass from the tail of needle to the tip during the warming treatment. The peak temperature on the body, tip of the silver needle in the multi-needle group is higher than those in the single needle group. Also, the peak temperature of multi-moxa ball is higher than that of single moxa ball.</p>

RESUMO

ObjectiveTo explore the effect of various forms of psychological intervention on psychological stress,mental state and treatment process of out- patients. Methods 198 outpatients from February 2010 to July 2011 were randomly divided into the observation group and the control group with 99 cases in each group.The control group received conventional treatment and care.The observation group was given multiple forms of psychological intervention on the basis of conventional treatment and care.Both groups were taken with Zung Anxiety Rating Scale (SAS)and the Hamilton Anxiety Scale(HAMA)to measure the psychological stress and the degree of anxiety before and after treatment.The total efficiencies of various disorders for the two groups before and after the diagnosis and treatment were compared. ResultsThere were no significant differences in SAS score and HAMA scores before psychological intervention.After the intervention,the SAS score and HAMA score in the control group were significantly higher than the observation group.The total effective rate after a course of treatment in the observation group was 90.6%.The total efficiency in the control group was 78.8%,which was significantly lower than that of the observation group. ConclusionsThe multiple forms of psychological intervention can reduce the mental stress,ease the patients' emotional state and improve the patients' therapy effect at the same period for out-patients.

<p><b>OBJECTIVE</b>To investigate the activation of mitogen-activated protein kinase (MAPK) pathway and the expression of inflammatory cytokine in lung tissue of rats with smoke inhalation injury, and to explore the injury mechanism.</p><p><b>METHODS</b>The model of smoke inhalation injury in airtight cabin was established. Thirty-six SD rats were randomly divided into normal control group (n = 6, NC group) and inhalation injury group (n = 30, II group). The rats in II group were observed at 1, 6, 24, 72 post injury hour (PIH) and 7 post injury day (PID). The pathological changes in lung tissue were observed by optical microscope. The contents of TNF-alpha, MIP-2, IL-1beta in lung tissue homogenate were examined. The level of p38MAPK, JNK, ERK1/2 and their phosphorylation in lung tissue were measured by Western blotting. The contents of TNF-alpha, MIP-2, and IL-1beta and granulocytes counts in BALF (bronchoalveolar lavage fluid) were detected by ELISA.</p><p><b>RESULTS</b>Pathological change of smoke inhalation was similar to acute lung injury. The contents of TNF-alpha and IL-1beta of lung tissue and BALF in II group at 1 PIH were higher than those in NC group (P < 0.01). The levels of MIP-2 of lung tissue in II group at each time point were similar to that in NC group (P > 0.05), while that of BALF in II group at 1 PIH were obviously higher than that in NC group (P < 0.01). Compared with those in NC group, the levels of p38MAPK, JNK, ERK1/2 in II group at each time point were similar, and their phosphorylation levels increased with different degree. Compared with that in NC group [(0.61 +/- 0.09) x 10(6) cells/L], the total number of granulocytes in II group was significantly decreased at 1PIH [(0.36 +/- 0.08) x 10(6) cells/L], increased on 7PID [(1.71 +/- 0.67) x 10(6) cells/L, P < 0.05]. The number of neutrophils in II group during 6PIH approximately 7 PID were more than that in NC group. The number of lymphocyte in II group at 1PIH less than that in NC group (P < 0.05), which increased gradually during 6 PIH approximately 7PID. There were no obvious difference in number of lymphocyte between two groups (P > 0.05).</p><p><b>CONCLUSION</b>The obvious inflammatory response in lung tissue can be induced by the poisonous gas released from the combustion of nonmetal materials in airtight cabin, and activate the expression of kinases in MAPK pathway, which may be one of mechanisms for lung injury induced by poisonous gas.</p>

<p><b>OBJECTIVE</b>To explore the molecular spectra and mechanism of human hypoxanthine guanine phosphoribosyl transferase (HPRT) gene mutation induced by ethyluitrosourea (ENU).</p><p><b>METHODS</b>Single cell cloning culture, two-way screening, multiple PCR amplification and electrophoresis technique were used.</p><p><b>RESULTS</b>With dose of ENU increasing, cell plating efficiency reduced (in the group with 100-200 micro g/ml doses, P < 0.01) and mutation frequency increased (in the group with 12.5-200.0 micro g/ml doses, P < 0.05) significantly. There was no all exons deletion in spontaneous mutations, and only 7.7% of them were detected as single exon deletion. But, deletion was found in 79.7% of ENU-induced mutations (62.5%-89.4%, P < 0.01), and deletion mutations in all nine exons of HPRT gene. Most of ENU-induced mutations were chain deletion with multiple exons (88.1%).</p><p><b>CONCLUSIONS</b>The spectra in spontaneous mutations differed completely from ENU-induced ones. ENU was liable to cause bigger changes in genetic structure, which suggested a stronger ENU's mutagenesis.</p>
